研究领域

低氧环境在高原和人类疾病(如实体瘤)中常常出现,低氧信号通路在肿瘤,血管生成等方面起着至关重要的作用。以往人们研究癌症,多收集癌组织和癌旁组织进行组学比较分析,获得差异表达基因信息,进而对差异候选基因进行体内外功能分析。本课题组利用基因组、转录组等进化生物信息学手段,比较分析高原家养动物及非模式动物在青藏高原迅速崛起的过程中低氧环境的适应机制,除开已知的HIF1/2信号通路以外,还筛选得到一系列有可能与低氧适应相关的候选基因,通过体外细胞存活、凋亡、肿瘤细胞的增殖、迁移及体内裸鼠荷瘤,Knock-In小鼠等实验方法证明,这些候选基因在肿瘤的发生发展过程中起着十分重要的功能。这些研究表明,利用进化学方法解析高原哺乳动物低氧适应的分子机理,同时能为人类疾病特别是实体瘤的研究、诊断或治疗提供新的靶点,提示交叉学科在未来人类疾病研究中具有极大的创新力及应用前景。
