研究领域

媒介昆虫传播的植物病毒病害对我国农业生产构成了严重威胁。深入解析植物的防御机制、昆虫的病原传递途径及病毒的致病机理,是当前植物保护研究的关键科学问题。在此研究基础上创制抗病种质是攻克农作物病虫灾害的有效策略。本研究组主要研究表观遗传信号在植物、昆虫和病毒的种内、种间的传递与调控机制,结合表观遗传学、基因组学、分子生物学和合成生物学等方法,挖掘植物与表观遗传相关的抗病基因并解析分子机理,最终将其用于农作物抗病种质的培育。

1.    表观遗传信号调控植物、昆虫与病毒三元互作

通过高通量测序技术,解析植物的转录组、翻译组、和表观基因组,包括DNA甲基化、组蛋白修饰(如乙酰化、甲基化)、染色质开放程度等,在植物与病虫害互作前后所发生的变化,挖掘植物潜在的抗病基因,并使用分子生物学与遗传学等方法进行验证。

2.    开发表观基因组编辑工具调控植物基因表达,创制抗病种质

CRISPR/dCas为基因靶标系统结合不同的表观遗传相关效应蛋白,开发出高效且精准的表观基因组编辑工具,通过改造目标位点的表观遗传修饰、调控基因表达。并将该工具运用于植物抗病基因的调控,创制抗病种质。
