研究领域

1. 蛋白质定向进化与新反应设计

天然酶促反应类型较为有限,如何突破原有反应类型,设计新酶促反应并用以合成新分子是本课题组研究的重要内容之一。酶定向进化技术是解决这一问题的有效手段,通过重构底物结合口袋的特定结合位点,改变原有的催化机制,赋予突变体新的催化反应;并对催化口袋微环境进行理性设计,控制突变体催化新反应的区域选择性及立体选择性。

2. 化学-酶法交叉合成新体系的设计与应用

通过将新的酶促反与有机合成相结合,对目标分子(活性天然产物分子和手性药物分子)发展绿色、高效、可持续的化学酶法交叉合成新路线。

3. 天然产物与手性药物分子的仿生半合成路线探索

通过微生物发酵方法以含有目标分子的基本骨架及其特定官能团的天然产物为起始原料,学习和借鉴天然产物生物合成途径中的关键元素,以增加合成的有效性和合理性;在合成战术层面,强调基于理性设计发展独具特色的合成方法,以提高合成的新颖性和实用性。
