致力于绘制面向脑科学与脑疾病研究的新一代人类脑图谱(脑网络组图谱),并建立其应用范式。聚焦特定认知功能的个体化脑图谱绘制,拟解决的关键科学问题包括:1. 如何利用机器学习、模式识别的方法,实现个体化脑图谱的构建;在此基础上,2. 如何解析特定认知功能的脑网络表征,为类脑智能研究提供借鉴?具体包括:利用多模态磁共振脑影像数据,发展个体化脑图谱绘制与验证的技术并构建可靠、稳定的个体化脑网络组图谱,并结合临床应用对图谱进行适用性评价;在此基础上,解析特定脑认知功能的脑网络表征,为理解脑的复杂功能提供帮助,进而为受脑科学知识启发的类脑智能研究提供借鉴。

1. 面向脑定位导航的个体化脑网络组图谱构建

随着以多模态磁共振成像技术为代表的神经影像学的迅速发展,以及与计算机后处理技术的紧密结合,衍生出立体定位辅助神经外科等新的理念,出现了脑立体定位导航系统等相关的技术。目前,无论是早期的有框架导航系统,还是近来迅速发展的无框架导航系统、以及脑外科辅助机器人,都已经在多种脑疾病的治疗上获得了重要的进展,拥有广阔的临床应用前景。由于不同类型脑疾病的病理机制差异巨大,不同患者之间的个体变异也十分显著,需要针对不同类型脑疾病以及患者,制定个性化的立体定向导航方案和针对性的神经调控策略。现有基于脑定位导航的神经调控治疗的瓶颈问题在于:缺乏精细尺度的脑图谱及其个体化临床解决方案,导致不能形成针对靶向目标的个性化精准治疗。亟需针对临床上不同类型脑疾病的多模态影像数据进行整合分析,优化、改进高精度个体化脑图谱的构建方法,绘制面向特定脑疾病神经调控治疗的个体化脑图谱。只有在此基础上,才能更有效的利用多模态脑成像技术研究不同因素对神经环路的动态影响,理解神经调控干预下的神经环路可塑性变化,从而明确脑疾病病理性改变的影像学特征,理解神经调控治疗的机制,为发现针对重大脑疾病早期诊断和预后评价的生物学标记提供基础。

2. 基于磁共振影像高等灵长类大脑联络皮层的跨物种比较研究

由于在神经系统结构和功能上,非人灵长类动物相较于其他实验动物与人类更为接近。因此,从神经系统进化的角度,开展灵长类近缘物种间的比较研究,进一步明确非人灵长类动物与人类在大脑联络皮层结构和功能上的异同,不但是了解人类大脑特有高级认知功能的一个重要突破口,同时也对建立重大脑疾病的非人灵长类动物模型,深入研究人类脑疾病的致病机制也具有重要的现实意义。跨物种比较是解析神经系统进化的基本工具,尤其是近年来比较神经影像学的快速发展,为跨物种比较提供了全新的视角和研究手段。我们拟利用高等灵长类 (人类、黑猩猩、猕猴与狨猴)的多模态磁共振脑影像数据,在不同物种大脑皮层个体化精细分区的基础上,定量分析大脑联络皮层的扩张程度及其连接模式的变化,建立物种之间同源脑区的对应关系,形成一套适用于跨物种比较的磁共振影像学研究框架,在宏观尺度上解析灵长类近缘物种间大脑皮层联络区进化上的异同。

今天的脑科学涵盖广泛,既有行为实验,又有功能性成像;既有细胞培养,又有数学建模;既有疾病研究,又有意识理论;既有动物实验,又有人类被试;既有计算机模拟,又有分子机理阐释;既有事实探究,又有工程发明……

如果你为了一腔年轻的热血放弃了赚大钱的机会,跑到冷门的基础学科学起了数学、物理、生物,然后发现学界的方向和自己的预想有那么一点点差距;或者你曾经为了赚大钱跑到热门的应用学科学起了计算机、医学、经济学,然后发现赚钱并不是自己的真正兴趣所在;又或者你曾经壮志满满的报考了工程系准备投身工业界,然后发现现实的需求不能满足自己发明的欲望,那么别气馁,至少你还有脑科学家这一条备胎。至少在我见过的同行中,他们的教育背景涵盖了上面列出的所有专业。至少我认为,交出志愿表的那一刻并不能决定人的一生。世界是圆的,转角永远都存在,好奇心也能带来比金钱更大的满足感。你想成为什么样的人,也许比想读什么专业更重要
