项目组情况

课题组于2008年11月成立,系统研究表观转录组调控与RNA甲基化功能机理,在拓展RNA甲基化表观转录组调控的前沿新领域合作研究中,取得了系列原创性发现,包括鉴定人源RNA m6A的修饰甲基转移酶复合物 METTL3/METTL14/WTAPm6A去甲基化酶ALKBH5/FTO Nat Chem Biol, 2011; Mol Cell, 2013; Cell Res, 2014a),揭示非编码miRNA介导的m6A甲基化位点形成的选择性机制(Cell Stem Cell,2015)及其m6A修饰的外显子被保留现象(Cell Res, 2014b)等。发现了m6A的识别因子(readerYTHDC1的的两个互作蛋白是mRNA剪接因子SRSF3SRSF10 Mol Cell2016RNA Biol, 2016)。揭示m5C调控mRNA出核的重要功能以及YTHDF3YTHDF1在翻译过程中的协同调控作用机制(Cell Res, 2017a, 2017b);,发现m6A通过YTHDF2介导notch1a mRNA的稳定性,维持内皮和造血细胞基因表达平衡,进而调控造血干细胞的命运决定(Nature, 2017;被评为2017中国生命科学十大研究进展);阐明了m6A调控脑发育和记忆作用机制(PLos Biol, 2018; Cell Res, 2018a,b)。对m6A的甲基转移酶、结合蛋白和去甲基化酶的生物学功能,及m6A在可变剪接等RNA代谢过程的调控作用进行了系统综述(Cell Res, 2018c)。
