研究领域

(1)新型纳微颗粒疫苗佐剂构建及其免疫增强机制研究。针对乙肝表面抗原构建了具有质子海绵效应的纳微颗粒,利用抗原的溶酶体逃逸提升细胞免疫功能,并基于此研发治疗性乙肝疫苗,进行免疫增强机制探索和治疗效果评价;针对HPV抗原,设计了具有pH敏感等相应释放的新型纳微颗粒,实现细胞和体液免疫的双重提升,与公司合作开发基于纳微颗粒的新型宫颈癌疫苗制剂;针对流感裂解抗原设计和构建了一系列新型纳微颗粒,开展相关免疫学评价,并研究免疫增强机制。同时,还系统研究了纳微颗粒理化性质对免疫学行为的影响,深化纳微颗粒佐剂设计。
(2)蛋白质药物微包埋中活性保持机制研究及其口服蛋白递送系统构建。针对微包埋过程中乳化过程、固化过程,系统研究了蛋白在微包埋过程中的失活原因,并提出了一系列的解决策略,实现了微包埋过程中蛋白多肽等活性药物的高活性保持,并针对口服蛋白多肽给药,设计构建了能够穿透粘液层的靶向杯状细胞的纳微颗粒口服给药体系,大大提高了药物的口服生物利用度;针对口服疫苗免疫效果不理想的问题,构建了具有M细胞靶向的脂质球递送系统,口服免疫获得了良好的效果。
(3)抗癌药物那为载体设计与制备研究。针对难溶抗肿瘤药物喜树碱等,设计和构建增加难溶药物溶出度及其具有靶向传输作用的纳微载体,研究抑瘤效果;同时,采用软模板技术,将哪容药物纳米化,大大提高了难溶药物的水溶性,而且可通过表面接枝进一步实现药物的靶向递送。
