Application of antibody array of splicing isoforms

It has been shown that most human genes are alternatively spliced and defects in alternative splicing (AS) contribute to cancer. However, currently, antibody microarrays are not used to study alternative splicing, which enhances proteome diversity and also modulate cancer-related proteins. Here we present an approach that allows the generation of an antibody microarray for the analysis of the expression of splice variants associated with cancer.

p73 isoforms and radiation-induced apoptosis

p73, has two distinct promoters, which allow the formation of two protein isoforms: full-length transactivating (TA) p73 and an N-terminally truncated p73 species (referred to as DNp73) that lacks the N-terminal transactivating domain. Our findings have shown that there is a differentialΔNp73 expression in response to different LET radiations, and downregulatedΔNp73 expression might play a critical role in sensitivity of tumor cells

★PCBP1 and p73 isoforms in cancer cells exposed to ionizing radiation

Based on the research ideal combined radiation and gene therapy, this project focused on the tumor hotspot of PCBP1as a bridge protein, has been found related to malignancy. PCBP1 expression regulates p73 isoforms expression and sensitizes cancer cells to ionizing radiationwhich is explored here. The aim of this project is to explore the radiation-induced tumor cell apoptosis molecular mechanism regulated by PCBP1.
