个人简介

鞠建华,男,1972年生,理学博士,博士生导师,中科院南海海洋研究所研究员,中国科学院大学岗位教授,广东省海洋药物重点实验室主任,中国科学院热带海洋生物资源与生态重点实验室主任。主要从事海洋微生物活性次级代谢产物的发现、生物合成和抗感染、抗肿瘤创新药物研发工作从海洋微生物中发现了具有抗感染、抗肿瘤等活性天然产物800余个,开发了海洋微生物的组合生物合成和异源表达技术,阐明了18种特征活性代谢产物的生物合成机制,揭示了咔啉碱合成酶、Dieckmann缩合酶、别异亮氨酸异构酶、细胞色素P450氧化酶、L-半乳糖变构酶等26种新颖生物合成酶的催化功能,筛选出3个自主产权的抗结核杆菌感染、抗胶质瘤和抗白血病候选海洋药物,其中1个在系统临床前研究主持国家重点研发计划、863计划重点课题、国家自然科学基金重点项目、国家海洋经济创新发展区域示范专项课题、广东省自然科学基金团队和中科院科技创新交叉团队项目等20余项。获得第五届施维雅青年药物化学奖(2002)、第七届药明康德生命化学研究奖(2013)。中国药学会海洋药物专业委员会副主任委员;中国微生物学会海洋微生物专业委员会副主任委员;热带海洋学报副主编;J. Nat. Prod.Mar. Drugs编委;中国海洋药物编委;广东药学会药物化学专业委员会副主任委员;广东海洋协会副会长/生物分会会长。2014年获得国家杰出青年科学基金。在Nat. Chem. Biol., Nature Commun., PNASJ. Am. Chem. Soc., Angew. Chem. Int. Ed., Org. lett., J. Nat. Prod.等学术刊物发表论文206篇,论文被SCI引用超过4300次,获授权专利22项。

研究领域:

本学科组以海洋微生物为研究对象,以海洋微生物活性次级代谢产物的生物学功能及其形成机制为拟解决的关键科学问题。主要从特殊海洋环境中(深海沉积物、珊瑚礁生态系统、不同深度的海水层、特色海洋生物等)分离、培养、鉴定海洋放线菌、真菌和细菌;综合运用微生物学、天然产物化学、细菌遗传学、分子生物学、生物信息学、生物化学和药理学等多学科专业技能,从海洋微生物中筛选发现新的生物活性物质,发掘新的生物合成途径、新型酶催化反应机理,利用代谢工程、组合生物合成和合成生物学技术手段构建新结构衍生物,并对具有自主产权、有前景的化合物进行成药性评价和药物开发,具体包括以下内容:

(1) 海洋微生物活性次级代谢产物的发现(Marine Bioactive Natural Products Discovery)。利用化学生态学原理和多种发酵培养技术,从海洋微生物中筛选、分离和鉴定结构新颖、活性显著的生物活性物质。研究海洋微生物产生的活性物质在特定海洋生态系统中的化学防御机理,发现生理活性显著药效活性物质,为开发具有我国独立知识产权的创新药物提供先导化合物。

(2) 海洋微生物复杂活性天然产物的代谢工程和组合生物合成(Metabolic Engineering and Combinatorial Biosynthesis of Complex Bioactive Natural Products)。包括:抗生素生物合成基因簇的克隆、序列测定和生物信息学分析;重要活性化合物产生菌全基因组的序列测定及其功能基因研究;新的生物合成途径的发现及其调控机制;基因阻断、置换、重组或异源表达构建工程菌,产生非天然的天然产物或提高目标天然产物的产量;利用基因克隆、蛋白表达、纯化手段,对酶促反应机理和动力学进行表征,发掘新型酶促反应催化剂;重要活性天然产物的体外全生物合成(in vitro total biosynthesis),体外构建重要天然产物的生物合成途径,用生物酶快速合成天然产物衍生物库。

(3) 抗感染、抗肿瘤海洋药物的成药性评价和药物研发(Anti-infective and Anti-tumor Drug Development)。对通过发现和生物合成技术改造获得的化合物进行构效关系研究,筛选出结构新颖、活性显著、具有自主知识产权的化合物进行体内药效、安全性、质量标准、药代和药剂学等临床前研究,开发抗感染、抗肿瘤海洋药物。

1. 学科组团队成员。研究员1, 马俊英(华南农业大学博士,分子生物学);副研究员3名,李青连(北京协和医学院博士,分子生物学),宋永相(中山大学博士,天然药物化学),孙长利(天然产物化学,中国科学院大学);助理研究员2名:秦湘静(中山大学博士,结构生物学),李骏(中国海洋大学博士,药学),在读博士生5名,硕士生7名,联合培养硕士生2名;已毕业博士生6名,硕士生10名。

2.硕士/博士研究生和博士后。招生专业一:海洋生物学(方向1:海洋微生物代谢工程、海洋生物技术;方向2:海洋微生物天然药物化学);招生专业二:生物工程。每年9月份接受教育部双一流高校,原985、211工程院校生命科学(生物科学、生物技术)、化学和药学专业推荐免试直博生和推荐免试硕士生各1名,接受联合培养硕士/博士研究生。
