Research Areas

Cell reprogramming (cellular reprogramming) refers to the process of reversion of cells to omnipotent or pluripotent state under specific conditions, or the formation of pluripotent stem cell lines, or further development into a new individual. Nuclear transfer and induced pluripotent stem cell (iPSC) technology are two main reprogramming methods. However, these two technologies are still immature. There are some problems such as low efficiency, poor reprogramming ability and poor reprogramming mechanism.


In 2012, the research team established the haploid embryonic stem cells from spermatozoa in mice and proved that these cells could replace sperm to make oocytes "fertilized" to produce semi cloned mice (called "semi cloned"). However, the semi cloning technique is not mature, the birth rate of half cloned mice is low (about 4%), and half of the half cloned mice have developmental defects. In 2015, the research team obtained the haploid stem cells (about 20%) of "spermatozoa", which could produce semoncloned mice, which could produce a semoncloned mouse, and proved that these cells could carry a large number of mice carrying different mutations with the CRISPR-Cas9 library, which laid the foundation for the genetic screening of the individual level of mice.


The laboratory studies the following aspects:


Objective to improve the method of cell reprogramming induced by nuclear transfer and iPSC technology, and to improve the efficiency of reprogramming.

1. to explore the molecular mechanism of reprogramming cell development defects and improve the quality of reprogramming cells.

2. to explore the molecular mechanisms of developmental defects in semi clonal mice, and to reveal important mechanisms related to embryonic development.

3. combining the haploid stem cells and CRISPR-Cas9 technology of "spermatozoa like cells", we will develop new gene editing applications.

4. carry out CRISPR-Cas9 mediated gene therapy research.
